Chirality is one of the basic attributes of nature. The study of chiral molecules is of great significance in the fields of life sciences, drug synthesis and asymmetric catalysis. The identification of the absolute configuration of chiral molecules is a challenging subject in the scientific community. Chiral Raman spectroscopy is a new spectroscopic method for the characterization of chiral molecules. Since this method does not require crystallization of the sample, it can directly identify the absolute configuration of the chiral sample in the solution phase, and is therefore subject to academia and industry. The community is highly concerned. However, the intrinsic signal of chiral Raman spectrum is very weak and is 3 to 7 orders of magnitude weaker than that of conventional spectroscopy. Therefore, it is very challenging to detect the chiral Raman signal experimentally. Based on years of research and development of UV Raman spectroscopy instruments, the research team proposed the development idea of ​​the short-wavelength chiral Raman spectroscopy instrument. Based on the analysis of the two basic principles of avoiding the electronic state absorption and avoiding the fluorescence interference, the optimized selection was suitable for the hand. The Raman spectroscopy 457nm laser as a light source, in cooperation with relevant domestic and foreign spectroscopic instrument companies, successfully developed the world's first short-wavelength chiral Raman spectrometer, which also fills the gap of our country's chiral Raman spectroscopy technology.
After the successful development of the instrument, a 457nm short-wavelength chiral Raman spectrum seminar was held at the 19th National Conference on Light Scattering. Six experts from domestic analytical chemistry, asymmetric synthesis, and drug research fields, experts in the field of Raman spectroscopy, and corporate technology leaders of chiral drugs such as the North China Pharmaceutical Group attended the meeting. Experts at the meeting held in-depth discussions after listening to the project manager Li Can’s summary reports, the development of other members of the project team, and the reports of on-site testing experts. At the end of the meeting, experts believed that the 457nm short-wavelength chiral Raman spectrometer was successfully developed for the first time in the world. The signal-to-noise ratio was greatly improved, the spectrophotometric time was reduced from hours to tens of minutes, and the sample to be tested was from pure compound to 10%. The performance of the chiral Raman instrument has reached a new height, and it also fills the technical gap in China's chiral Raman spectroscopy instrument. It is also proposed that the short-wavelength chiral Raman spectrometer be engineered as soon as possible. The spectrometer is expected to play an important role in the identification of chiral molecules, synthesis and identification of new drugs, asymmetric catalysis, and research of biological macromolecules.
