Chlorophyll meter to determine the conditions of the extract

The chlorophyll extraction conditions in red clover pretreatment wastewater were studied by chlorophyll meter determination and analysis of single factor and orthogonal experiments. The results showed that the optimal ethanol concentration for the preparation of crude chlorophyll extract from chlorophyll deposited in wastewater was 30%. The optimum extraction conditions of chlorophyll in crude extract were ethanol concentration 90%, extraction temperature 60°C, extraction time 6 h, and solid-liquid ratio 1:25 (g/mL).

Under this condition, the extraction rate of chlorophyll can reach 1.72mg/g. According to the results of orthogonal experiments, the effect of these four factors on the extraction rate of chlorophyll was ethanol concentration> solid-liquid ratio> extraction temperature> extraction time. The chlorophyll measured by chlorophyll meter has good heat resistance and oxidation resistance, and is stable in weak acid and weak alkali environments. Fe2+ contributes to its stability, but it is prone to degradation under light conditions.

The oxidant has an effect on the stability of the chlorophyll, but it is not obvious. With the increase of hydrogen peroxide concentration, although the content is reduced, but the magnitude is not large, indicating that the chlorophyll antioxidant capacity is strong. The chlorophyll meter has found that various metal ions have different effects on the stability of chlorophyll, and Fe2+ contributes to its stability improvement and produces a good color protection effect; Zu2+ and Al2+ have little effect on its stability, but Cu2+ And Fe2+ accelerates the decomposition of chlorophyll, which is not conducive to its time preservation.

The chlorophyll meter experiment adopts the alcohol precipitation method to settle the organic matter in the red clover pretreatment wastewater, and uses the alcohol extraction method to extract the chlorophyll in the sediment. The operation is simple, the titer ratio is high, the reagent used is safe and nontoxic, and the chlorophyll loss rate is small. It provides a scientific basis for the purification and effective use of this type of wastewater and similar wastewater. The resulting red clover chlorophyll has potential application value in the food, pharmaceutical and other industries.
